Uncertain significance for Arrhythmogenic cardiomyopathy with wooly hair and keratoderma; Arrhythmogenic right ventricular dysplasia 8 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004415.4(DSP):c.7750A>G (p.Ser2584Gly), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the DSP gene (transcript NM_004415.4) at coding-DNA position 7750, where A is replaced by G; at the protein level this means replaces serine at residue 2584 with glycine — a missense variant. Submitter rationale: This sequence change replaces serine, which is neutral and polar, with glycine, which is neutral and non-polar, at codon 2584 of the DSP protein (p.Ser2584Gly). This variant is present in population databases (rs755017000, gnomAD 0.0009%). This variant has not been reported in the literature in individuals affected with DSP-related conditions. An algorithm developed to predict the effect of missense changes on protein structure and function outputs the following: PolyPhen-2: "Benign". The glycine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
