This variant lies in the BRIP1 gene (transcript NM_032043.3) at 3 bases into the intron immediately before coding-DNA position 1795, where T is replaced by G. Submitter rationale: This sequence change falls in intron 12 of the BRIP1 gene. It does not directly change the encoded amino acid sequence of the BRIP1 protein. RNA analysis indicates that this variant induces altered splicing and may result in an absent or altered protein product. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with BRIP1-related conditions. Variants that disrupt the consensus splice site are a relatively common cause of aberrant splicing (PMID: 17576681, 9536098).
