Pathogenic for STAT3 gain of function; Hyper-IgE recurrent infection syndrome 1, autosomal dominant — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_139276.3(STAT3):c.2137G>C (p.Val713Leu), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces valine, which is neutral and non-polar, with leucine, which is neutral and non-polar, at codon 713 of the STAT3 protein (p.Val713Leu). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with Hyper IgE syndrome (PMID: 18602572, 25962528; Invitae). In at least one individual the variant was observed to be de novo. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt STAT3 protein function with a positive predictive value of 80%. For these reasons, this variant has been classified as Pathogenic.
