Uncertain significance for Combined oxidative phosphorylation defect type 7; Spastic paraplegia — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_152269.5(MTRFR):c.202C>A (p.His68Asn), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces histidine, which is basic and polar, with asparagine, which is neutral and polar, at codon 68 of the C12orf65 protein (p.His68Asn). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with C12orf65-related conditions. An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
