This variant lies in the FBN1 gene (transcript NM_000138.5) at coding-DNA position 8497 through coding-DNA position 8498, deleting 2 bases; at the protein level this means shifts the reading frame starting at serine residue 2833, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This sequence change creates a premature translational stop signal (p.Ser2833Tyrfs*5) in the FBN1 gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 39 amino acid(s) of the FBN1 protein. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with FBN1-related conditions. This variant disrupts a region of the FBN1 protein in which other variant(s) (p.Met2864Lys) have been determined to be pathogenic (Invitae). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing.
