This variant lies in the PLEC gene (transcript NM_201384.3) at coding-DNA position 13566, where C is replaced by A; at the protein level this means converts the codon for tyrosine at residue 4522 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: This sequence change creates a premature translational stop signal (p.Tyr4549*) in the PLEC gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 26 amino acid(s) of the PLEC protein.
