This variant lies in the SQSTM1 gene (transcript NM_003900.5) at coding-DNA position 1160, where C is replaced by A; at the protein level this means replaces proline at residue 387 with glutamine — a missense variant. Submitter rationale: This sequence change replaces proline, which is neutral and non-polar, with glutamine, which is neutral and polar, at codon 387 of the SQSTM1 protein (p.Pro387Gln). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with SQSTM1-related conditions. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed at Invitae for this missense variant, however the output from this modeling did not meet the statistical confidence thresholds required to predict the impact of this variant on SQSTM1 protein function.
