NM_001111.5(ADAR):c.244A>G (p.Ser82Gly) was classified as Uncertain significance for Aicardi-Goutieres syndrome 6; Symmetrical dyschromatosis of extremities by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces serine, which is neutral and polar, with glycine, which is neutral and non-polar, at codon 82 of the ADAR protein (p.Ser82Gly). This variant is present in population databases (no rsID available, gnomAD 0.006%). This variant has not been reported in the literature in individuals affected with ADAR-related conditions. An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
