Uncertain significance for Inclusion body myopathy with Paget disease of bone and frontotemporal dementia; Frontotemporal dementia and/or amyotrophic lateral sclerosis 6 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_007126.5(VCP):c.115G>A (p.Val39Met), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the VCP gene (transcript NM_007126.5) at coding-DNA position 115, where G is replaced by A; at the protein level this means replaces valine at residue 39 with methionine — a missense variant. Submitter rationale: This sequence change replaces valine, which is neutral and non-polar, with methionine, which is neutral and non-polar, at codon 39 of the VCP protein (p.Val39Met). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with VCP-related conditions. An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
