Uncertain significance for Charcot-Marie-Tooth disease dominant intermediate E; Focal segmental glomerulosclerosis 5 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_022489.4(INF2):c.313G>A (p.Val105Met), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces valine, which is neutral and non-polar, with methionine, which is neutral and non-polar, at codon 105 of the INF2 protein (p.Val105Met). The frequency data for this variant in the population databases is considered unreliable, as metrics indicate poor data quality at this position in the gnomAD database. This variant has not been reported in the literature in individuals affected with INF2-related conditions. Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed for this missense variant. However, the output from this modeling did not meet the statistical confidence thresholds required to predict the impact of this variant on INF2 protein function. This variant disrupts the p.Val105 amino acid residue in INF2. Other variant(s) that disrupt this residue have been observed in individuals with INF2-related conditions (PMID: 30373780; internal case), which suggests that this may be a clinically significant amino acid residue.
