Uncertain significance for Hypertrophic cardiomyopathy 25; Primary familial hypertrophic cardiomyopathy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003673.4(TCAP):c.376C>T (p.Gln126Ter), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the TCAP gene (transcript NM_003673.4) at coding-DNA position 376, where C is replaced by T; at the protein level this means converts the codon for glutamine at residue 126 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: This sequence change creates a premature translational stop signal (p.Gln126*) in the TCAP gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 42 amino acid(s) of the TCAP protein. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with TCAP-related conditions. Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
