This variant lies in the PLEC gene (transcript NM_201384.3) at coding-DNA position 1522 through coding-DNA position 1523, deleting 2 bases; at the protein level this means shifts the reading frame starting at leucine residue 508, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This sequence change creates a premature translational stop signal (p.Leu535Alafs*35) in the PLEC gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in PLEC are known to be pathogenic (PMID: 20301336, 20447487, 21109228, 23289980, 28824526). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with PLEC-related conditions.
