Uncertain significance for 46 XY differences of sex development; Oligosynaptic infertility — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004959.5(NR5A1):c.962G>T (p.Gly321Val), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glycine, which is neutral and non-polar, with valine, which is neutral and non-polar, at codon 321 of the NR5A1 protein (p.Gly321Val). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individuals with NR5A1-related conditions (PMID: 33351340; Invitae). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt NR5A1 protein function with a negative predictive value of 80%. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
