Uncertain significance for Hypertrophic cardiomyopathy 12; Dilated cardiomyopathy 1M — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003476.5(CSRP3):c.536C>A (p.Thr179Lys), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the CSRP3 gene (transcript NM_003476.5) at coding-DNA position 536, where C is replaced by A; at the protein level this means replaces threonine at residue 179 with lysine — a missense variant. Submitter rationale: This sequence change replaces threonine, which is neutral and polar, with lysine, which is basic and polar, at codon 179 of the CSRP3 protein (p.Thr179Lys). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with CSRP3-related conditions. An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
