Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Color Diagnostics, LLC DBA Color Health to NM_007294.4(BRCA1):c.2702T>C (p.Phe901Ser), citing ACMG Guidelines, 2015. This variant lies in the BRCA1 gene (transcript NM_007294.4) at coding-DNA position 2702, where T is replaced by C; at the protein level this means replaces phenylalanine at residue 901 with serine — a missense variant. Submitter rationale: This missense variant replaces phenylalanine with serine at codon 901 of the BRCA1 protein. Computational prediction suggests that this variant may not impact protein structure and function (internally defined REVEL score threshold <= 0.5, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. In a large breast cancer case-control study, this variant has been observed in 2/60466 cases and 1/53461 controls (PMID: 33471991). This variant has been identified in 1/250872 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
