Submitter rationale: This sequence change replaces methionine, which is neutral and non-polar, with threonine, which is neutral and polar, at codon 340 of the AIFM1 protein (p.Met340Thr). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individuals with clinical features of Charcot-Marie-Tooth disease (PMID: 25590979, 28967629, 31523922).
