This variant lies in the CBS gene (transcript NM_000071.3) at coding-DNA position 844, where C is replaced by T; at the protein level this means replaces proline at residue 282 with serine — a missense variant. Submitter rationale: A novel variant of uncertain significance has been identified in the CBS gene. The P282S variant has not been published as a pathogenic variant, nor has it been reported as a benign variant to our knowledge. This variant was not observed in approximately 6,500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. The P282S variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. This substitution occurs at a position that is conserved across species, and in silico analysis predicts this variant is probably damaging to the protein structure/function. However, to our knowledge no studies have been performed to determine the functional effect of the P282S variant.Therefore, based on the currently available information, it is unclear whether this variant is pathogenic or benign.
