NM_007129.5(ZIC2):c.1076-5C>T was classified as Uncertain significance by GeneDx, citing GeneDx Variant Classification (06012015): The c.1076-5C>T variant in the ZIC2 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. The c.1076-5C>T variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). This variant has been identified as a de novo variant with confirmed parentage. However, in-silico analyses, including splice predictors and evolutionary conservation, suggest that this variant does not alter protein structure/function. Therefore, we interpret c.1076-5C>T as a variant of uncertain significance.
