Uncertain significance — the classification assigned by GeneDx to NM_006393.3(NEBL):c.2718C>A (p.Ser906Arg), citing GeneDx Variant Classification (06012015). This variant lies in the NEBL gene (transcript NM_006393.3) at coding-DNA position 2718, where C is replaced by A; at the protein level this means replaces serine at residue 906 with arginine — a missense variant. Submitter rationale: A variant of uncertain significance has been identified in the NEBL gene. The S906R variant has not been published as a pathogenic variant, nor has it been reported as a benign variant to our knowledge. This variant was not observed in approximately 6,500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. The S906R variant is a semi-conservative amino acid substitution, which may impact secondary protein structure as these residues differ in some properties. However, this substitution occurs at a position where amino acids with similar properties to Serine are tolerated across species and in silico analysis is inconsistent in its predictions as to whether or not the variant is damaging to the protein structure/function.Therefore, based on the currently available information, it is unclear whether this variant is pathogenic or rare benign. This result cannot be interpreted for diagnosis or used for family member screening at this time.
