This variant lies in the PIK3CD gene (transcript NM_005026.5) at coding-DNA position 3029, where A is replaced by C; at the protein level this means replaces glutamic acid at residue 1010 with alanine — a missense variant. Submitter rationale: The E1010A variant in the PIK3CD gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The E1010A variant was not observed at any significant frequency in approximately 6500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. The E1010A variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. This substitution occurs at a position where amino acids with similar properties to Glutamic Acid are tolerated across species. In silico analysis predicts this variant is probably damaging to the protein structure/function. We interpret E1010A as a variant of uncertain significance.
