Likely oncogenic for Acute myeloid leukemia — the classification assigned by Molecular Diagnostics Division, Virginia Commonwealth University to NM_001127208.3(TET2):c.3678dup (p.Val1227fs), citing ClinGen/CGC/VICC Guidelines for Oncogenicity, 2022 erratum: This variant was not observed in gnomAD or somatic databases. The variant is predicted to lead to a truncation of the protein which results in the loss of all of the catalytic domain (Tet_JBP, source PFAM) which encompasses amino acids 1290 to 1905 of the 2002 amino acid protein. Catalytic activity of TET2 is required for suppression of myeloid malignancy [PMID: 27003514]. This variant is therefore a typical null variant in a tumor suppressor and is not present in population databases which renders it to be at minimum likely oncogenic.
