NM_006295.3(VARS1):c.646C>G (p.Leu216Val) was classified as Uncertain significance for Neurodevelopmental disorder with microcephaly, seizures, and cortical atrophy by Neuberg Centre For Genomic Medicine, NCGM, citing ACMG Guidelines, 2015: The observed missense variant c.646C>G(p.Leu216Val) in the VARS1 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This variant is absent in the gnomAD Exomes. The amino acid Leu at position 216 is changed to a Val changing protein sequence and it might alter its composition and physico-chemical properties. Multiple lines of computational evidence (Polyphen - Benign, SIFT - Tolerated and MutationTaster - Polymorphism) predict no damaging effect on protein structure and function for this variant. The residue is conserved by GERP++ and PhyloP across 100 vertebrates. For these reasons, this variant has been classified as Uncertain Significance. Another significant variant in this gene has been detected in the spouse.
