NM_002069.6(GNAI1):c.302C>T (p.Ala101Val) was classified as Uncertain significance for Neurodevelopmental disorder with hypotonia, impaired speech, and behavioral abnormalities by Neuberg Centre For Genomic Medicine, NCGM, citing ACMG Guidelines, 2015: The observed missense variant c.302C>T(p.Ala101Val) in the GNAI1 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This variant is absent in the gnomAD Exomes. The amino acid Ala at position 101 is changed to a Val changing protein sequence and it might alter its composition and physico-chemical properties. Computational evidence (Polyphen - Benign, SIFT - Tolerated and MutationTaster - Disease causing) predicts conflicting evidence on protein structure and function for this variant. The residue is conserved by GERP++ and PhyloP across 100 vertebrates. For these reasons, this variant has been classified as Uncertain Significance.
