This variant lies in the SETD1A gene (transcript NM_014712.3) at coding-DNA position 932, where G is replaced by C; at the protein level this means replaces arginine at residue 311 with proline — a missense variant. Submitter rationale: The missense variant c.932G>C (p.Arg311Pro) in the SETD1A gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This variant is absent in the gnomAD Exomes. The amino acid Arg at position 311 is changed to a Pro changing protein sequence and it might alter its composition and physico-chemical properties. Multiple lines of computational evidence (Polyphen - Damaging, SIFT - Damaging and MutationTaster - Disease causing) predict a damaging effect on protein structure and function for this variant. The amino acid change p.Arg311Pro in SETD1A is predicted as conserved by GERP++ and PhyloP across 100 vertebrates. For these reasons, this variant has been classified as Uncertain Significance
