NM_033310.3(KCNK4):c.317A>T (p.Tyr106Phe) was classified as Uncertain significance for Facial dysmorphism, hypertrichosis, epilepsy, intellectual/developmental delay, and gingival overgrowth syndrome by Neuberg Centre For Genomic Medicine, NCGM, citing ACMG Guidelines, 2015: The observed missense variant c.317A>T(p.Tyr106Phe) in the KCNK4 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This variant is absent in the gnomAD Exomes. The amino acid Tyr at position 106 is changed to a Phe changing protein sequence and it might alter its composition and physico-chemical properties. Multiple lines of computational evidence (Polyphen - Damaging and MutationTaster - Disease causing) predict a damaging effect on protein structure and function for this variant. The residue is conserved by GERP++ and PhyloP across 100 vertebrates. For these reasons, this variant has been classified as Uncertain Significance.
