This variant lies in the AUTS2 gene (transcript NM_015570.4) at coding-DNA position 1298, deleting one base; at the protein level this means shifts the reading frame starting at leucine residue 433, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: The c.1298delT pathogenic variant in the AUTS2 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The c.1298delT variant causes a frameshift starting with codon Leucine 433, changes this amino acid to a Proline residue, and creates a premature Stop codon at position 40 of the new reading frame, denoted p.Leu433ProfsX40. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The c.1298delT variant was not observed in approximately 6,500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. We interpret c.1298delT as a pathogenic variant.
