Pathogenic — the classification assigned by GeneDx to NM_001365536.1(SCN9A):c.2141G>A (p.Trp714Ter), citing GeneDx Variant Classification (06012015). This variant lies in the SCN9A gene (transcript NM_001365536.1) at coding-DNA position 2141, where G is replaced by A; at the protein level this means converts the codon for tryptophan at residue 714 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: The W703X pathogenic variant in the SCN9A gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The W703X variant was not observed in approximately 5900 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. We interpret W703X as a pathogenic variant.
