NM_001291415.2(KDM6A):c.2356A>G (p.Thr786Ala) was classified as Likely pathogenic by GeneDx, citing GeneDx Variant Classification (06012015): The T734A variant in the KDM6A gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The T734A variant was not observed in approximately 6500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. The T734A variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. This substitution occurs at a position that is not conserved. In silico analysis predicts this variant likely does not alter the protein structure/function. The T734A variant is a strong candidate for a pathogenic variant.
