Uncertain significance for Hereditary sensory and autonomic neuropathy type 1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_006415.4(SPTLC1):c.277dup (p.Thr93fs), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the SPTLC1 gene (transcript NM_006415.4) at coding-DNA position 277, duplicating one base; at the protein level this means shifts the reading frame starting at threonine residue 93, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This sequence change creates a premature translational stop signal (p.Thr93Asnfs*15) in the SPTLC1 gene. It is expected to result in an absent or disrupted protein product. However, the current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in SPTLC1 cause disease. This variant is present in population databases (rs746304060, gnomAD 0.0009%). This variant has not been reported in the literature in individuals affected with SPTLC1-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
