This variant lies in the PLEC gene (transcript NM_201384.3) at coding-DNA position 7531, where C is replaced by T; at the protein level this means converts the codon for glutamine at residue 2511 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: The Q2538X pathogenic variant in the PLEC gene has been reported previously in association with EB with pyloric atresia in a neonate (Nakamura et al., 2005). This variant is predicted to cause loss of normal protein function either through protein truncation, as the last 2,037 amino acids are lost. The Q2538X variant was not observed in approximately 6,500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. We interpret Q2538X as a pathogenic variant.
