Uncertain significance for Congenital myasthenic syndrome 18 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_130811.4(SNAP25):c.12C>G (p.Asp4Glu), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces aspartic acid, which is acidic and polar, with glutamic acid, which is acidic and polar, at codon 4 of the SNAP25 protein (p.Asp4Glu). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with SNAP25-related conditions. An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated.
