NM_013322.3(SNX10):c.303C>G (p.Phe101Leu) was classified as Uncertain significance by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces phenylalanine, which is neutral and non-polar, with leucine, which is neutral and non-polar, at codon 101 of the SNX10 protein (p.Phe101Leu). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with osteopetrosis (internal data). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
