Pathogenic — the classification assigned by GeneDx to NM_000444.6(PHEX):c.883del (p.Ala295fs), citing GeneDx Variant Classification (06012015): The c.883delG pathogenic variant in the PHEX gene causes a frameshift starting with codon Alanine 295, changes this amino acid to a Proline residue and creates a premature Stop codon at position 6 of the new reading frame, denoted p.Ala295ProfsX6. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. Although this variant has not been previously reported to our knowledge, we interpret is to be pathogenic.
