Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001039591.3(USP9X):c.3538G>A (p.Gly1180Ser), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glycine, which is neutral and non-polar, with serine, which is neutral and polar, at codon 1180 of the USP9X protein (p.Gly1180Ser). This variant is present in population databases (no rsID available, gnomAD 0.002%), including at least one homozygous and/or hemizygous individual. This variant has not been reported in the literature in individuals affected with USP9X-related conditions. An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
