This variant lies in the IL12RB1 gene (transcript NM_005535.3) at the canonical splice donor site of the intron immediately after coding-DNA position 580, where G is replaced by A; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: This sequence change affects a donor splice site in intron 6 of the IL12RB1 gene. It is expected to disrupt RNA splicing. Variants that disrupt the donor or acceptor splice site typically lead to a loss of protein function (PMID: 16199547), and loss-of-function variants in IL12RB1 are known to be pathogenic (PMID: 9603733, 12591909). This variant is not present in population databases (gnomAD no frequency). Disruption of this splice site has been observed in individual(s) with IL12RB1-related conditions (PMID: 21057261).
