Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant allele was found at a frequency of 4.2e-06 in 236296 control chromosomes. c.427A>T has been observed in two individuals affected with Canavan Disease (Zeng_2006). These data indicate that the variant may be associated with disease. A different variant affecting the same codon has been classified as likely pathogenic/pathogenic by our lab (c.428T>C, p.Ile143Thr), supporting the critical relevance of codon 143 to ASPA protein function. At least one publication reports experimental evidence evaluating an impact on protein function (Zano_2013). The most pronounced variant effect results in only residual enzyme activity.
