This variant lies in the TBK1 gene (transcript NM_013254.4) at coding-DNA position 140, where G is replaced by A; at the protein level this means replaces arginine at residue 47 with histidine — a missense variant. Submitter rationale: This sequence change replaces arginine, which is basic and polar, with histidine, which is basic and polar, at codon 47 of the TBK1 protein (p.Arg47His). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with amyotrophic lateral sclerosis (PMID: 25803835). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is not expected to disrupt TBK1 protein function with a negative predictive value of 95%.
