Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001987.5(ETV6):c.109G>A (p.Val37Met), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces valine, which is neutral and non-polar, with methionine, which is neutral and non-polar, at codon 37 of the ETV6 protein (p.Val37Met). This variant is present in population databases (rs752186257, gnomAD 0.002%). This missense change has been observed in individual(s) with leukemia (PMID: 26102509). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is not expected to disrupt ETV6 protein function with a negative predictive value of 80%. Experimental studies have shown that this missense change does not substantially affect ETV6 function (PMID: 26102509). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
