Submitter rationale: This sequence change replaces methionine, which is neutral and non-polar, with isoleucine, which is neutral and non-polar, at codon 645 of the MYO7A protein (p.Met645Ile). RNA analysis indicates that this missense change induces altered splicing and likely results in a shortened protein product. This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individuals with autosomal recessive MYO7A-related conditions (PMID: 21031134, 33576163).
