Uncertain significance for Mosaic variegated aneuploidy syndrome 2 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_014679.5(CEP57):c.699G>A (p.Glu233=), citing Invitae Variant Classification Sherloc (09022015): This sequence change affects codon 233 of the CEP57 mRNA. It is a 'silent' change, meaning that it does not change the encoded amino acid sequence of the CEP57 protein. This variant also falls at the last nucleotide of exon 6, which is part of the consensus splice site for this exon. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with CEP57-related conditions. Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown. Variants that disrupt the consensus splice site are a relatively common cause of aberrant splicing (PMID: 17576681, 9536098).
