Pathogenic for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_032043.3(BRIP1):c.205+1del, citing Ambry Variant Classification Scheme 2023: The c.205+1delG intronic pathogenic mutation, located in intron 2 of the BRIP1 gene, results from a deletion of one nucleotide within intron 2 of the BRIP1 gene. This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). Alterations that disrupt the canonical splice site are expected to cause aberrant splicing, resulting in an abnormal protein or a transcript that is subject to nonsense-mediated mRNA decay. RNA studies have demonstrated that this alteration results in abnormal splicing in the set of samples tested (Ambry internal data). As such, this alteration is interpreted as a disease-causing mutation.
