NM_001613.4(ACTA2):c.479G>A (p.Gly160Asp) was classified as Uncertain significance for Aortic aneurysm, familial thoracic 6 by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glycine, which is neutral and non-polar, with aspartic acid, which is acidic and polar, at codon 160 of the ACTA2 protein (p.Gly160Asp). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individuals with clinical features of thoracic aortic aneurysm and dissection (PMID: 19409525; Invitae). It has also been observed to segregate with disease in related individuals. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt ACTA2 protein function with a negative predictive value of 80%.
