Uncertain significance for Multiple endocrine neoplasia, type 2 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_020975.6(RET):c.2299A>C (p.Ser767Arg), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces serine, which is neutral and polar, with arginine, which is basic and polar, at codon 767 of the RET protein (p.Ser767Arg). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with Hirschprung disease (PMID: 7581377). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. Experimental studies have shown that this missense change affects RET function (PMID: 10921886, 11438491, 14715928, 22837065). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
