This variant lies in the PRKAG2 gene (transcript NM_016203.4) at coding-DNA position 997, where T is replaced by C; at the protein level this means replaces serine at residue 333 with proline — a missense variant. Submitter rationale: This sequence change replaces serine, which is neutral and polar, with proline, which is neutral and non-polar, at codon 333 of the PRKAG2 protein (p.Ser333Pro). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with hypertrophic cardiomyopathy (PMID: 28431061). It has also been observed to segregate with disease in related individuals. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed at Invitae for this missense variant, however the output from this modeling did not meet the statistical confidence thresholds required to predict the impact of this variant on PRKAG2 protein function.
