This variant lies in the REEP1 gene (transcript NM_001371279.1) at coding-DNA position 104 through coding-DNA position 105, deleting 2 bases; at the protein level this means shifts the reading frame starting at tyrosine residue 35, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This sequence change creates a premature translational stop signal (p.Tyr35Cysfs*32) in the REEP1 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in REEP1 are known to be pathogenic (PMID: 18321925, 18644145, 32655478). This variant is not present in population databases (gnomAD no frequency). This premature translational stop signal has been observed in individual(s) with autosomal dominant hereditary spastic paraplegia (PMID: 18321925). It has also been observed to segregate with disease in related individuals. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site.
