Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000374.5(UROD):c.659A>C (p.His220Pro), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces histidine, which is basic and polar, with proline, which is neutral and non-polar, at codon 220 of the UROD protein (p.His220Pro). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with hepatoerythropoietic porphyria (PMID: 8176248, 22382040). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. This variant is also known as c.677A>C. ClinVar contains an entry for this variant (Variation ID: 3720201). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed for this missense variant. However, the output from this modeling did not meet the statistical confidence thresholds required to predict the impact of this variant on UROD protein function. Experimental studies have shown that this missense change affects UROD function (PMID: 8176248). For these reasons, this variant has been classified as Pathogenic.
