Uncertain significance — the classification assigned by Ambry Genetics to NM_013266.4(CTNNA3):c.189G>T (p.Glu63Asp), citing Ambry Variant Classification Scheme 2023: The p.E63D variant (also known as c.189G>T), located in coding exon 2 of the CTNNA3 gene, results from a G to T substitution at nucleotide position 189. The glutamic acid at codon 63 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
