Uncertain significance for TWIST1-related craniosynostosis — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_006494.4(ERF):c.137T>C (p.Ile46Thr), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces isoleucine, which is neutral and non-polar, with threonine, which is neutral and polar, at codon 46 of the ERF protein (p.Ile46Thr). This variant is present in population databases (rs769908132, gnomAD 0.0009%). This variant has not been reported in the literature in individuals affected with ERF-related conditions. Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is expected to disrupt ERF protein function with a positive predictive value of 95%. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
