Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001220.5(CAMK2B):c.1877T>C (p.Ile626Thr), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces isoleucine, which is neutral and non-polar, with threonine, which is neutral and polar, at codon 626 of the CAMK2B protein (p.Ile626Thr). This variant is present in population databases (rs771083993, gnomAD 0.002%). This variant has not been reported in the literature in individuals affected with CAMK2B-related conditions. An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant¬†is likely to be tolerated.
