NM_000179.3(MSH6):c.3215_3222del (p.Gly1072fs) was classified as Pathogenic for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.3215_3222delGTCCTATG pathogenic mutation, located in coding exon 5 of the MSH6 gene, results from a deletion of 8 nucleotides at nucleotide positions 3215 to 3222, causing a translational frameshift with a predicted alternate stop codon (p.G1072Vfs*18). This variant has been detected in a patient with colon cancer that demonstrated high microsatellite instability and loss of MSH6 staining by immunohistochemistry (Ambry internal data). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
